Achievements:
Important progress on the
basic research of novel drug delivery systems based on the nanotechnology has
been achieved, including:
1. Employing endogenous transport mechanism to design targeted drug delivery
systems: applying multiple receptors and transport pathways with tumor-targeted
and high blood-brain-barrier crossing transport capacity, we evaluate their high
efficiency mediated brain-targeted transport and the mechanism. Meanwhile we
found that compared with antibody or protein with high molecular weight, small
targeted molecules have higher fabricability and efficiency, which make them
with better potential in drug development and clinical applications. Furthermore,
based on the characteristic that blood-brain-barrier and glioma cells endow the
same target, we select the choline & glucose derivate, which can target
both blood-brain-barrier and glioma, to realize the encephalic drug
dual-targeted focus, and to decrease the influence to the normal brain parts
and peripheral tissues.
2. Design of microenvironment triggered targeted therapy strategy to realize
targeted delivery and focused release: in line with the pathema’s physiology
and pathology microenvironment characteristics, we construct the tissue/cell microenvironment
triggered and controlled release system to modulate the drug focused release,
which will provide theoretical references for the design of high efficiency and
low toxic brain-targeted formulation.
3. In line with the pathema’s physiology and pathology characteristics, we
design, synthesize and optimize polymer materials, which can “on-demand” fit
the requirement for the drug or therapy ", to construct the carriers with
highly drug-carrying, controlled release, biodegradable, low toxic and side-effect
capacity. We mainly focused on the amino acid-based polymers.
4. Employing the nanotechnology based on the dendritic macromolecules and chitosan
as carriers to increase the oral availability of insoluble drugs.
5. We have one PhD candidate winning the “Excellent Doctor Degree Dissertation
of Shanghai City” and three master candidates wining “Excellent Master Degree Dissertation
of Shanghai City”.
